FDA Approves Rasonque (daraxonrasib), Ushering in a New Era of Targeted Pancreatic Cancer Therapy
On August 26, 2026, the U.S. Food and Drug Administration (FDA) granted expedited approval to Rasonque (daraxonrasib), a first-of-its-kind daily oral RAS inhibitor for adults with metastatic pancreatic adenocarcinoma. The FDA approved the drug 6.5 months ahead of the scheduled user fee deadline, following Breakthrough Therapy and Orphan Drug designations. The agency had previously permitted an expanded access program in May 2026 due to the drug's high anticipation and therapeutic promise.
Clinical Data and Trial Design
The approval is backed by a randomized, open-label, multicenter Phase III clinical trial involving 500 adults with previously treated metastatic pancreatic adenocarcinoma whose tumors carried specific cancer-driving mutations (predominantly KRAS):
- Doubled Survival: Patients receiving Rasonque achieved a median overall survival of 13.2 months, compared to 6.7 months for those receiving standard chemotherapy.
- Mechanism of Action: Daraxonrasib works as a "molecular glue" that binds to and blocks mutated proteins in the RAS gene family, which regulates cell growth and fuels tumor development in more than 90% of pancreatic cancer cases.
- Tolerability: The drug demonstrated significantly fewer severe side effects than standard chemotherapy regimens.
Access and Commercialization
Revolution Medicines announced that the daily pill will carry a list price of approximately $39,800 for a one-month supply. While not a cure, oncologists emphasize that Rasonque represents a paradigm shift that opens the door to targeting KRAS mutations across other difficult-to-treat tumor types, including lung cancer.
