One-Shot CRISPR Cholesterol Therapy CTX310 Holds Its LDL Cuts at One Year
A first-in-human Phase 1 trial of CTX310 — a one-time CRISPR-Cas9 infusion that switches off the liver gene ANGPTL3 — has shown its lipid-lowering effect persisting a full year, according to Cleveland Clinic researchers presenting at the 2026 European Society of Cardiology annual meeting and publishing simultaneously in the New England Journal of Medicine (Laffin et al., DOI 10.1056/NEJMc2609825).
The data. The trial followed 15 patients with lipid disorders that had not responded adequately to medications, extending the two-month results first reported in November 2025. "After 12 months, participants who received the highest dose had a 52.5% reduction from baseline in LDL cholesterol and a 47.8% drop in triglycerides." Doses ranged from 0.1 to 0.8 mg/kg, pre-treated with corticosteroids and antihistamines. "No serious adverse events related to the therapy were reported during the one-year follow-up." Patients will be followed for another 15 years, per FDA recommendations for gene-editing therapies. The study was funded by CRISPR Therapeutics AG.
First author Luke Laffin (Cleveland Clinic): "Building upon the initial data presented in November 2025, the durability of the lipid-lowering effect was impressive... It is encouraging that there were no serious safety events related to CTX310 in the trial and in the year following treatment. We look forward to continuing to investigate this therapy in a larger number of patients."
What it means. Durability is the central bet for "one-and-done" gene editing against cardiovascular risk — a single infusion that keeps LDL near-halved at 12 months, with no serious related safety events, is the strongest evidence yet that the concept survives past the initial months1. Keep the caveats: n=15, no cardiovascular-outcomes data, and ANGPTL3 editing must clear larger trials. Watch for the Phase 2 program and whether competing gene-editing lipid programs report comparable durability.
